Abstract

Ischemic tissue injury represents a leading cause of global morbidity and mortality, necessitating advanced therapeutic interventions aimed at promoting functional recovery. A critical determinant of successful tissue repair is the dynamic regulation of macrophage polarization, a process wherein macrophages transition from an initial pro-inflammatory phenotype to a subsequent pro-resolving and regenerative state. Traditional studies have predominantly relied on single-layer biological analyses, which often fail to capture the complex, interconnected regulatory networks governing this phenotypic plasticity. This paper presents a comprehensive examination of macrophage polarization in ischemic tissue repair through a multi-omics lens, integrating transcriptomics, epigenomics, proteomics, and metabolomics. By synthesizing data across these molecular strata, we elucidate the profound metabolic reprogramming events, particularly the shifts between glycolysis and oxidative phosphorylation, and their direct mechanistic links to epigenetic landscape remodeling. The analysis underscores how these multi-dimensional datasets can identify novel regulatory hubs and potential therapeutic targets that are entirely obscured in single-omics paradigms. Furthermore, we discuss the methodological challenges and computational strategies required to harmonize heterogeneous datasets. Ultimately, the integration of systems biology into macrophage immunology provides a robust framework for designing next-generation immunomodulatory therapies for ischemic cardiovascular and peripheral vascular diseases
