Abstract

The paradigm of oncological treatment has increasingly shifted from systemic, non-specific chemotherapy to localized, highly targeted interventions. Among the numerous biomaterials investigated for localized drug delivery, chitosan-based hydrogels have emerged as exceptionally versatile platforms capable of both stimuli-responsive drug release and active immunomodulation. This paper provides a comprehensive academic analysis of the synthesis, mechanisms, and therapeutic applications of smart chitosan hydrogels in cancer therapy. By capitalizing on the mildly acidic, hyper-metabolic, and reductive nature of the tumor microenvironment, these intelligent biopolymer networks can be engineered to release chemotherapeutic agents with precise spatiotemporal control. Furthermore, the inherent cationic properties of chitosan serve a dual purpose, acting not merely as an inert structural scaffold but as a potent immunoadjuvant. The material demonstrates a profound ability to reprogram immunosuppressive tumor microenvironments by facilitating the maturation of dendritic cells and promoting the polarization of macrophages toward an anti-tumor phenotype. Through a detailed examination of contemporary literature, methodological synthesis strategies, and aggregated analytical results, this paper highlights the synergistic potential of combining responsive drug delivery with localized immunotherapy. The discussion also addresses the mechanical, degradative, and biocompatibility profiles of these systems, paving the way for future clinical translation in oncology.
